User Related Lists
Show all records, workflow alerts, dashboards, etc. related to and/or owned by a given User in Related Lists under the User's record.
This would greatly aid in User management, as you'd be able to see all the records, rules, etc., that need to be managed when activating and deactivating users. Likewise, users could see in one place all the records they own or are affiliated with via lookup fields. This would also greatly expland User-related reporting.
News Feeds
News Feeds are a great way to keep up to date with your team as well as learn about other things that are happening in your organization. Living on your home page, it's a quick sampling of recent activity around the records that interest you, and a jumping off point to take action.
Here is some feedback we've heard from customers:
“A lot of times our lenders change the opportunity but you don't know it until you go in and view it, so I think this is really helpful. I would want to know if the account owner changed, or a contact was added to a company."
“The constant workflow items that come up: if a case meets a certain criteria, or a contact... or if a new asset is created, sales announcements, opportunities closing, new products. If it were customizable enough, we could push certain fields, kind of like a workflow trigger."

A couple people point out in the comments that an in-app reader is great, but for those who use blog readers, RSS/Atom subscriptions will be important as well.

Salesforce Social
I believe Salesforce.com should harness the power of Social Networking.
By allowing a user to add "team members" to their team list (either internal or extral via Salesforce to Salesforce).
Then a section on the home page should be used to describe updates from their team, like facebook.
By adding relevant and timely information about a users's teams activities we can drive adoption, best practices and collaboration!
Dashboards: better control over size of components
It would be great if we could have more control over the appearance of dashboard components.
Examples: I would like to be able to
define a base height and width, so that similar components all have the same size,
let "high" components span multiple rows,
let "wide" components span multiple columns,
add labels/titles that can span multiple components (this we can probably do with embedded s-controls, but it would be grand to have a simpler way)
